• Screening of putative positive promoter caulimovirus clones by use of miniprep DNA isolation,miniprep DNA digestion and subsequently separation of fragments using agarose gel electrophoresis..
• Preparation of competent cells using TB1 culture and subsequently carrying out transformation, followed by restriction digestion and agarose gel electrophoresis.
• Ligation of foreign DNA into the caulimovirus vector.
• Elution of DNA from agarose gel and subsequent ligation and cross checking with help of restriction digestion.
• Assembly and Rescue PCR techniques..
