equence specific DNA-binding transcription factor and these two proteins p63 and p73 which share extensive homology to p53 family with different Isoforms. Our group tried to unravel the molecular transcriptional mechanism by which the various members and its isoforms of p53 family cross talk and influence the fate of the cells. The tumor suppressor function of the p53 protein occurs via its function as a transcription factor which regulates gene expression.

My specific contribution in the project were:
· Screening the colonies of different tumor suppressor genes and its isoforms p53, p63 and p73.
· Transposition of the fastback (clones) into Bacmids and transfecting the DNA into Baculovirus expression system (Eukaryotic expression system) which is an excellent expression system because of its post translational modifications.
· Insect cells are infected with virus which we get after transfection.
· Purification of Flag tagged and His tagged protein by various column chromatography and affinity chromatography methods using different antibodies (resins).
· The protein is checked by various techniques as PAGE analysis, Western analysis and Gel Shift.
Insect cell lines SF9 and SF21 used for baculovirus expression system and mammalian cell lines as Hela cells, MCF-7,MDA-MB 231, DU-145 (different cervical and breast cancer cell lines) are under study.
MTT assay and nuclear extract preparation of these cell lines are done.

Wet lab Skills:

DNA and RNA related work:
PCR(Polymerase chain Reaction), Plasmid based cloning, Restriction analysis of plasmid DNA , Plasmid isolation in small and large scale using minipreps and midipreps, Agarose Gel Electrophoresis, Isolation of Genomic DNA , Isolation of mRNA , Northern analysis, Southern blotting, Primer designing.

Protein work:
SDS PAGE, Western PAGE, Gel shift Analysis, Purification of proteins by affinity chromatography and estimation of proteins and amino acids.

Spectrophotometric techniques:
UV Spectrophotometer, DNA estimation, RNA estimation, Protein estimation.

Flow Cytometry
FACS – Florescence assisted cell sorting of different cancer cell lines to study the effect of anticancer drugs.

Nuclear extract preparation- of different cervical and prostrate cancer cell lines.
MTT assay- To check the effect of different anticancer drug on different cervical and prostrate cancer cell lines.

Microbial work:
Bacterial transformation (with various E.coli cells DH10 beta, DH5 etc)
Transposition (using DH10 Bacmid)
Agrobacterium mediated transformation.

Animal cell culture:
Worked on Baculovirus expression system (Eukaryotic expression system) using insect cell culture like SF9 and SF21 cell lines. Mammalian cell culture involving HeLa cell lines, cervical and breast cancer cell lines like MCF-7 and MDMB468 etc.

Plant tissue culture:
Inoculation of seeds in M.S media
Regeneration in B1N1 media
Raising of transgenics by introducing different constructs using Agrobacterium mediated transformation using competent cells.
