Topic: - Studies on microbial lipases- Isolation and screening of cultures, enzyme production, Purification and characterization.
Isolation and screening work done at Dept. of Biotechnology, Guru Jambheshwar University of Science & Technology, Hisar, Haryana under Guidance of project supervisor Dr. Neeraj Dilbaghi (Reader, Dept. of Bio and Nanotechnology, GJUST, Hisar). Further studies including enzyme production, purification, characterization and modification of lipase was done at Enzyme Lab., Dept. of Biochemical Engineering & Biotechnology, Indian Institute of Technology-Delhi, Hauz Khas under Guidance of project co- supervisor Dr. Subhash Chand (Professor, DBEB, IIT- Delhi)

2) M.Sc training programme at Regional Research Laboratory, CSIR, Jammu
Duration: - 5 weeks
Topic: - Production and characterization of lipases produced by bacterial strains.
Description: Strains producing lipases were screened; quantitative estimation and growth profile were studied for the selected isolates. Fermentation was performed in STR (7 L fermentor) followed by characterization.
Under Guidance of project guide Dr. Rajinder Parshad (Head, Pilot plant, RRL, CSIR) and project supervisor Dr. Raina (Senior Scientist, Pilot plant, CSIR, Jammu).

Description- Isolation and screening of microorganisms possessing lipolytic activity was studied and characterization of Pseudomonas aeruginosa was done with respect to industrial properties.
2) Praveen Dahiya, Subhash Chand., Ashok Chaudhury & Neeraj Dilbaghi. 2008. Lipase production by Pseudomonas mendocina M-37 in batch & continuous culture. J. Agri. Biol. Res., 24(1):18-29.
Description- Lipase production by Pseudomonas mendocina M-37 and growth were investigated in submerged culture under batch fermentation and the yield & productivity were compared to that of continuous mode of fermentation using STR.
3) Dilbaghi, N., Dahiya, P., Chaudhury, A. and Subhash Chand. Characterization of an extracellular alkaline lipase from Pseudomonas mendocina M-37. Paper communicated to Braz. J. Microbiol. (Status- Amendment requested)
Description- A potent strain of Pseudomonas mendocina producing extracellular lipase was isolated from soil. This lipase was capable of hydrolyzing a variety of lipidic substrates and is mainly active towards synthetic triglycerides and fatty acid esters that possess a butyryl group. Metal ions like Mg2+, Ca2+ and Na+ stimulated lipase activity.
4) “Production and purification of an extracellular lipase from Pseudomonas mendocina M-37”- Research paper communicated to Bioresource Technology.
Description- The possibilities of increasing lipase production by Pseudomonas strain with different growth conditions was investigated. The appropriate source of nitrogen and carbon and other physical parameters involved were tested for further lipase yield. We succeeded in increasing the maximum lipolytic activity approx. 3.7 fold and specific activity 4.4 fold over the initial valves obtained with the non- optimized medium.
5) “Isolation and screening of extracellular lipase production by Pseudomonas mendocina M-37 and its characterization”- Paper presented at the 8th national symposium, BIOHORIZON-2006 at IIT- Delhi
Description- Certain kinetic characteristics of the Pseudomonas mendocina lipase enzyme isolated from soil sample and its stability in organic solvents was studied. The characterization of the Pseudomonas mendocina lipase showed that it was thermostable, pH stable, stable in various organic solvents and exhibiting 1,3 regiospecificity.
6) “Lipase production by isolated Pseudomonas sp. M-54 in Batch and Continuous culture” - Paper presented at 7th national symposium, BIOHORIZON-2005, IIT- Delhi
Description- Using the optimized conditions, lipase production and growth were investigated in submerged culture of Pseudomonas mendocina M-54 under batch fermentation and the yield & productivity were compared to that of continuous mode of fermentation using stirred tank reactors.
7) “Isolation of lactose fermenting yeast strain for ethanol production from lactose & waste from dairy processing industries”- paper presented at the national symposium in Emerging Environmental Issues and Technological Challenges, Dept. of Environmental Science & Engineering, G J University, Hisar.
Description- Ethanol production using lactose and waste from dairy processing industry was studied. Various nutritional and physical conditions were optimized in order to increase the ethanol production.
